理化学研究所などの共同研究グループは、微小試験管アレイを用いた1分子計測技術を基盤とする全自動デジタルバイオ分析装置の製品化プロトタイプを開発した。装置には、高精度な自動分注、加熱・磁気操作、小型蛍光イメージング、高速画像解析などを統合し、試料の前処理から試薬混合、反応、画像取得、データ解析、結果表示までを自動化した。約50cm四方のベンチトップ型で、さまざまなデジタルバイオ分析法に対応できる汎用プラットフォームとして設計されている。感染症検査では、鼻咽頭ぬぐい液を直接投入して約15分で結果を得られ、SATORI法による急性呼吸器感染症検査で感度・特異度とも約90%を実証した。熟練者への依存を減らし、測定時間やばらつきを抑えられるため、感染症の迅速診断に加え、がんや認知症の早期診断、さらに農業・畜産・環境分野への応用も期待される。

Summary

Digital bioanalysis enables single-molecule detection of biomolecules and holds transformative potential across the life sciences and clinical diagnostics. However, its broader translation remains limited by accessibility and versatility. Here, we present COWFISH2A+, a compact, fully automated device integrating wide-field imaging with a custom dispensing module. Using an open-type microchip, COWFISH2A+ enables diverse assays at low cost through direct liquid deposition. As a proof of concept, we demonstrate two types of digital bioanalysis: a digital enzyme assay and an amplification-free digital genetic assay, SATORI. The digital enzyme assay discriminates alkaline phosphatase, a blood biomarker, at the single-isozyme level with sub-femtomolar sensitivity, while SATORI achieves attomolar-level detection of multiple pathogen-derived genetic targets. Furthermore, to demonstrate the translational potential of SATORI, we directly test 159 clinical specimens, detecting SARS-CoV-2 from nasopharyngeal swabs within ∼20 min with 92% sensitivity and 85% specificity. COWFISH2A+ thus provides an accessible and versatile platform for digital bioanalysis.
