Abstract
Advanced paternal age is a recognized risk factor for autism spectrum disorder (ASD), yet the molecular mechanisms linking paternal aging to early neurodevelopment remain poorly understood. Although ASD exhibits marked sex differences in prevalence and clinical presentation, whether paternal aging exerts sex-dependent effects on the embryonic brain has not been fully examined. Here, we generated a bulk RNA-sequencing dataset from embryonic day 14.5 mouse telencephalon derived from young (3-month-old) and aged (19-month-old) fathers. To minimize variability arising from developmental asynchrony, embryonic stages were rigorously matched using a hindlimb-based morphological staging method. Principal component analysis and hierarchical clustering revealed transcriptomic variation associated with both paternal age and sex. Notably, transcriptomic profiles of female aged father-derived embryos formed a distinct cluster and showed a tendency toward increased gene expression, suggesting a sex-biased transcriptional response to paternal aging. This dataset provides a well-controlled transcriptomic resource for investigating how paternal aging influences early brain development in a sex-dependent manner.
